研究方向

DNA修補機轉

隨著分子生物學的快速進展及發現,癌化乃是由於染色體或基因遭到損害而造成細胞不正常的生長。其過程常牽涉了整個基因之改變,其中基因之功能損傷與修補扮演一非常重要之角色。諸多報告顯示先天DNA修補基因缺陷之病人易造成口腔癌化,但DNA斷裂缺損修補基因的變異是否和口腔癌的發生有關及其機轉,仍有待深入研究。過去已用小鼠基因剔除術 (gene knockout) 模式探討其可能機轉,現更進一步嚐試印證其在臨床病人之表現。

 訊號傳遞

生物體經由正常之細胞內代謝都會產生一些活潑性氧分子,如過氧化氫、超氧自由基、氫氧自由基。根據統計,在此正常 狀況下,人體每個細胞每天大約會發生10,000 個DNA的鹼基損害。此外,細胞 暴露於外來因子諸如離子輻射、紫外線、或環境中之毒素更會增加基因變異的發生。這些傷害可能造成細胞生長停滯或細胞凋亡,但目前對其調控機制之訊號傳遞瞭解仍有許多疑問有待釐清,本實驗室目前對於matriptase及IEX-1於毒害物質及藥物處理下所造成的訊號傳遞變化已頗有進展。
